I'm no assassin but I do have a some knowledge of the Dark Brotherhood. Higher ranking members communicate with assassins using what they call "dead drops." They are hidden places where they can place instructions about contracts, so they never have to meet face to face and risk being seen. I happen to know where one is. You can use it to get the letter to the Brotherhood.

On the road between Anvil and Kvatch, there's a place called the Gottshaw Inn. Go to the stable. There, one of the hay bales has a hollowed-out spot in it. That's where they hide messages. Just put the letter in there and the Brotherhood will get it....and try not to disturb anything!
